A parthenogenetic Artemia population from Torre Colimena, southernItaly, originally reported in 1998, is characterized by a multidisciplinaryapproach including cyst and naupliar biometry, morphometry ofadults, scanning electron microscopy (SEM) of brood pouch, cytogeneticsand 16S rRNA PCR-RFLP analysis. We confirmed parthenogeneticstatus, inferred ploidy level and determined phenotypic andmolecular relationships of this population through comparisonswith other asexual Artemia strains as well as bisexual speciesfound (A. salina) or introduced (A. franciscana) in the Mediterraneanbasin. Cyst and naupliar sizes for Torre Colimena are amongthe smallest recorded for asexual Artemia while the oppositeis true for chorion thickness. Discriminant analysis of adultbody measurements shows increased differentiation (89.5% forthe first four out of the twelve functions produced) from tetraploidparthenogenetic strains and bisexual species (A. salina andA. franciscana). Scanning electron micrographs of brood pouchreveal the characteristic morphology of asexual strains, whilechromosome observations of instar-I nauplii unequivocally establishdiploidy. Restriction patterns give evidence that the TorreColimena population shares an identical set of mitochondrialDNA (mtDNA) polymorphisms with a diploid Spanish parthenogeneticstrain and it is well differentiated from other tetraploid aswell as bisexual auto- and allochthonous strains (A. salinaand A. franciscana) from the Mediterranean. As a collaborative work of three laboratories the polymorphism of the canine fourth complement component (C4) was studied in a total of 131 unrelated dogs from different breeds and mongrels. Using high voltage electrophoresis followed by an immunoblotting technique, we detected eight distinct variants. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of canine C4 showed an additional heterogeneity of the and chains which resulted in a total of 11 variants in the population studied. So that more precise information concerning the respective C4 allotypes will be available, a nomenclature is proposed designating not only the migration pattern of the C4 variants in agarose gels but also the heterogeneity of the C4 chains observed in SDS-PAGE.Abbreviations used in this paper AGE

Considering that quality water supplies are diminishing and climate disorder affects water cycle, wastewaters should be decontaminated for reuse either by the same establishment or in agriculture for the growth of industrial plants. In that context, much research work has been focused on the development of low cost biosorbents. In this study, the effect of composting on the adsorption capacity of olive tree pruning waste (OTPW) biomass for methylene blue (MB) removal from aqueous solutions was examined. Composting procedure may improve the sorption properties of the raw organic materials, is economical and easy to apply. MB adsorption on both OTPW and composted olive tree pruning waste (COTPW) biomasses was found to be fast. The maximum monolayer adsorption capacity obtained from Langmuir isotherm was estimated to be 129.87 and 250.00 mg/g for OTPW and COTPW, respectively, indicating that composting procedure greatly improved the adsorptive properties of OTPW. The raise of temperature from 25°C to 60°C decreased the efficiency of OTPW for MB removal whereas the adsorption capacity of COTPW was not affected at high temperatures. A novel nanocomposite cathode consisting of sulfur and hollow‐mesoporous titania (HMT) embedded within carbon nanotubes (CNT), which is designated as S‐HMT@CNT, has been obtained by encapsulating elemental sulfur into the pores of hollow‐mesoporous, spherical TiO2 particles that are connected via CNT. A carbon‐paper interlayer, referred to as dual functional porous carbon wall (DF‐PCW), has been obtained by filling the voids in TiO2 spheres with carbon and then etching the TiO2 template with a chemical process. The DF‐PCW interlayer provides a medium for scavenging the lithium polysulfides and suppressing them from diffusing to the anode side when it is inserted between the sulfur cathode and the separator. Lithium–sulfur cells fabricated with the thus prepared S‐HMT@CNT cathode and the DF‐PCW interlayer exhibit superior performance due to the containment of sulfur in TiO2 and improved lithium–ion and electron transports. The Li–S cells display high capacity with excellent capacity retention at rates as high as 1C, 2C, and 5C rates. 相似文献

Experimental sepsis can be induced in mice using the cecal ligation and puncture (CLP) method, which causes polymicrobial sepsis. Here, a protocol is provided to induce sepsis of varying severity in mice using the CLP technique. Autophagy is a fundamental tissue response to stress and pathogen invasion. Two current protocols to assess autophagy in vivo in the context of experimental sepsis are also presented here. (I) Transgenic mice expressing green fluorescence protein (GFP)-LC3 fusion protein are subjected to CLP. Localized enhancement of GFP signal (puncta), as assayed either by immunohistochemical or confocal assays, can be used to detect enhanced autophagosome formation and, thus, altered activation of the autophagy pathway. (II) Enhanced autophagic vacuole (autophagosome) formation per unit tissue area (as a marker of autophagy stimulation) can be quantified using electron microscopy. The study of autophagic responses to sepsis is a critical component of understanding the mechanisms by which tissues respond to infection. Research findings in this area may ultimately contribute towards understanding the pathogenesis of sepsis, which represents a major problem in critical care medicine. 相似文献

Environmental and host-associated microbial communities are complex ecosystems, of which many members are still unknown. Hence, it is challenging to study community dynamics and important to create model systems of reduced complexity that mimic major community functions. Therefore, we developed MiMiC, a computational approach for data-driven design of simplified communities from shotgun metagenomes. We first built a comprehensive database of species-level bacterial and archaeal genomes (n = 22 627) consisting of binary (presence/absence) vectors of protein families (Pfam = 17 929). MiMiC predicts the composition of minimal consortia using an iterative scoring system based on maximal match-to-mismatch ratios between this database and the Pfam binary vector of any input metagenome. Pfam vectorization retained enough resolution to distinguish metagenomic profiles between six environmental and host-derived microbial communities (n = 937). The calculated number of species per minimal community ranged between 5 and 11, with MiMiC selected communities better recapitulating the functional repertoire of the original samples than randomly selected species. The inferred minimal communities retained habitat-specific features and were substantially different from communities consisting of most abundant members. The use of a mixture of known microbes revealed the ability to select 23 of 25 target species from the entire genome database. Several studies showed signs of autonomic dysfunction in patients with primary Sj?gren's syndrome (pSS). Adrenomedullary function might be of importance for pSS pathogenesis by affecting salivary gland functions and modulating immune responses. The aim of the study was to evaluate the adrenomedullary hormonal system in patients with pSS. The glucagon test (1 mg i.v.) was performed in 18 pSS patients and 13 control subjects. During the test each patient had electrocardiographic and impedance cardiographic monitoring. Plasma epinephrine and norepinephrine were assayed by liquid chromatography with electrochemical detection after batch alumina extraction. Baseline concentrations of epinephrine and norepinephrine were comparable between pSS and controls. Glucagon administration induced a significant increase in systolic blood pressure, diastolic blood pressure, heart rate, cardiac output (P < 0.01), and stroke volume; however, the changes were comparable between pSS and controls. Epinephrine levels increased (P < 0.01) in response to glucagon administration while norepinephrine concentration did not change. There was no significant difference in neurochemical responses to glucagon between pSS and controls. In conclusion, the present results suggest normal adrenomedullary function in pSS. 相似文献
